How do I exclude specific contacts from a group before sending to that list?

I'd suggest using a Segment. You can pull everyone from your group and exclude certain folks based upon their email addresses. Here's more information on Segments.

Just chiming in here to second Kyle's suggestion to use the segmenting tool to isolate those addresses you'd like to exclude from your group. The easiest way to do this would be to segment your audience by audience group, response history or contact information to isolate just the contacts from that group you do want to send to. Then you can send to that segment instead of the original group! You'll find more detailed instructions on segmenting in the resource article linked above.

Thanks for your replies.  I'm still a bit confused though.  When I create a segment, it looks just like a group.  And, when I try to exclue someone from the segment (so that they don't receive the email), by deleting them, it only gives me the option of deleting them entirely from my audience.  I don't want to delete them entirely, I simply just want to exclude them from this particular email.
Any advice would be great.

Hey WA44 -- I see where the confusion is here. Let's think of the resulting segment as the list youwant to send to. The addresses you want to exclude should be excluded during the segmenting process.

So, let's say you typically send to Group A, but this time you'd like to exclude anyone who has never opened an email. You could build a segment based on groups and response history to find anyone in Group A thathasopened an email. That resulting segment would be the folks from Group A that you want to send to, excluding those that you wanted to leave out of this send. Does that help to clarify things a bit?

Tell me a little bit more about who you're trying to exclude and based on what information, and I can walk you through building that segment. I will say, that if you're just looking to exclude a few specific contacts from a group, it might be simpler to copy the entire group over and delete those contacts from the group (not your entire audience). Let me know how I can help!
Emily - I am having a similar issue. I do not want to exclude them based on any particular past behavior, I just have a list of names tat should NOT receive this specific mailing. Is there a way to exclude individuals from a segment?

Hi St. Andrew's -- There isn't currently a way to exclude contacts from a segment other than to make sure they no longer meet the criteria that segment is searching by. If there isn't a way to alter the related data in the contact records to eliminate those contacts from the segment, you'll need to do some group work.

Here's what I would do - manually add each of those contacts to a new group (Group A). Next, right before sending (so that it's up to date), copy your segment to another group (Group B). Then, run a new segment to find anyone who exists in Group B but not in Group A, and send to that resulting list. 

Since your exclusion list doesn't really have any commonalities, they'll be some manual effort here. I hope this helps!
I ended up creating a segmented list that included the two large groups, then importing that segment into a new group, and manually deleting the contacts that needed to be deleted.
